For whatever reason...
For me, it's Audrey Tautou's title character in 'Amélie', Katherine Hepburn in 'Bringing Up Baby', Drew Barrymore in 'The Wedding Singer'.
You?

I'd have to say Vesper Lind (Eva Green) from Casino Royale ... I loved how she matched wits with James Bond and put up a substantial challenge for his bullet-proof bravado. She was still able to break through all his defenses and charm and still fall for him (rather than regard him as an inferior).
Less of a heroine, but still lovable, was Audrey Farmer (Virginia Madsen) in The Astronaut Farmer. She tried to snap her husband out of his "fantasy", but she realized she was mistaken and encouraged him to fulfill his dreams.
Battlestar Galactica has two heroines I love - President Laura Roslin (Mary McDonnell) and Sharon "Athena" Agathon (Grace Park).

Bryce Dallas Howard in 'The Village'. She's the best part of the movie.
Scarlett Johannsen in 'Lost in Translation'. I knew how Bill Murray felt.
Deborah Foreman in 'Valley Girl'. Ah, early 80's big-hair chicks.

I agree with Drew Barrymore in The Wedding Singer
Naomi Watts - Mullholland Drive
Jennifer Connelly - A Beautiful Mind
Renée Zellweger - Chicago
Not necessarily falling in love, but these particular characters made them very like-able.

Sally Albright - When Harry Met Sally...
Kathleen Kelly - You've Got Mail
Hmmm... come to think of it, maybe I've just got a thing for Meg Ryan.
Slightly off topic... Lorelai Gilmore. I know she's a TV character, but what the hell!

I wouldn't say I fell in love with him, but James Gandolfini as Tony Soprano was amazing. The guy's a thug, seriously bad news, just plain mean in some episodes, yet you still had to feel for him doing what he could to put food on the table for his family. That's quality writing, when you cheer on the bad guy.
